人胰腺癌细胞系MIA PaCa-Ⅱ神经侵袭原位移植瘤动物模型的建立 被引量:1

Establishment of animal model of in situ transplanted tumor of human pancreatic cancer MIA PaCa Ⅱ cells with activity of nervous invasion

摘要 目的建立人胰腺癌细胞系MIA PaCa-Ⅱ裸鼠胰腺原位移植瘤模型并观察神经侵袭情况,为研究胰腺癌神经侵袭的发生、发展提供实验依据和条件。方法将人胰腺癌细胞系MIA PaCa-Ⅱ行裸鼠背部皮下接种,建立高转移特性胰腺癌裸鼠皮下移植瘤模型,然后将高转移性移植瘤组织接种于裸鼠胰腺被膜下,分别于4周、6周和8周处死裸鼠行移植瘤组织解剖和病理学检查,测量移植瘤大小和重量,并用HE和银染方法观察神经侵袭情况。结果模型建立过程中,胰腺癌细胞系MIA PaCa—Ⅱ细胞裸鼠皮下接种成瘤率为100%(10/10),皮下移植瘤呈局限性生长,无脏器和神经转移。原位移植4周、6周和8周后,胰腺癌原位移植瘤成瘤率均为100%(10/10),神经转移率分别为50%(5/10)、80%(8/10)和60%(6/10),并可见多个脏器转移,以肝脏、肝门淋巴结、胃窦转移和腹膜播散多见。结论成功建立了人胰腺癌细胞系MIA PaCa-Ⅱ裸鼠胰腺原位移植瘤模型并观察了神经侵袭情况,此模型为一较理想的“拟人”神经浸润转移模型,可用于胰腺癌体内嗜神经性机制的研究。 Objective To establish the nude mice model of in situ transplanted tumor of human pancreatic cancer MIA PaCa Ⅱ cells and observe its activity of nervous invasion to provide experimental basis and condition for study the occurrence and development of the nervous invasion by pancreatic cancer. Methods The human pancreatic cancer MIA PaCa Ⅱ cells were implanted on back of nude mice to establish the subcutaneous model of transplanted tumor of pancreatic cancer with high activity of metastasis in nude mice. Then the tumor was implanted in situ in the pancreas of the nude mice. The nude mice was sacrificed in 4, 6 and 8 weeks for anatomy and pathological examination of the transplanted tumor as well as determination of its size and weight. Furthermore, HE staining and silver staining were employed to observe its activity of nervous invasion. Results During the establishment of subcutaneous model, the successful rate of implantation was 100% (10/10). The subcutaneous transplanted tumor had limited growth and there was no metastasis to organs and nerves. In 4, 6 and 8 weeks after the in situ pancreatic implantation, the successful implantation rate was 100% (10/ 10). The rate of nervous invasion was 50% (5/10), 80% (8/10) and 60% (6/10), respectively. Meanwhile, there were metastases of the tumor in various organs. Conclusion The in situ transplanted tumor model was successfully established in nude mice and its nervous invasion observed. This model is a fairly optimal model for study of nervous invasion by pancreatic cancer.
