摘要 [目的]探索五种罗非鱼的亲缘关系。[方法]从尼罗罗非鱼、奥利亚罗非鱼、杂交种尼奥罗非鱼、莫桑比克罗非鱼和红罗非鱼基因组中提取DNA,通过PCR扩增其ITS1基因序列及18S、5.8S部分序列并进行序列分析。[结果]扩增片段大小约700 bp,测序结果显示获得的序列包括完整的ITS1区以及18S和5.8S部分序列。序列长度变异主要在ITS1区,18S和5.8S片段的序列长度没有发生变化。罗非鱼5个种的ITS1序列的碱基组成差别不大。从UPGMA聚类结果上看,尼罗罗非鱼与尼奥罗非鱼聚为一支;莫桑比克罗非鱼与奥利亚罗非鱼聚为一支,红罗非鱼再与莫-奥聚合。[结论]五种罗非鱼的ITS1及18S、5.8S部分序列分析结果表明,尼罗和尼奥罗非鱼亲缘关系较近,奥利亚与莫桑比克罗非鱼亲缘关系较近,红罗非鱼可能与奥利亚和莫桑比克罗非鱼有一定的亲缘关系。 [ Objective ] The aim is to understand the genetic relationship among five species Oreochromis Tilapias as well as to provide interspecifie molecular markers for breeding practice. [ Method ] The genomic DNA of Oreochromis niloticus, O. aureus ,hybrids Ni-ao (O. niloticus × O. aureus) ,O. mossambicus and red tilapia were extracted, and their ITS1 gene sequences were amplified by PCR and analyzed. [ Result ] The amplified segment was about 700 bp and the sequencing result showed that the obtained sequence included complete ITS1 domain and partial sequence of 18S and 5.8S. The sequence length variation was mainly in the ITS1 domain and the sequence length of 18S and 5.8S segments had no change. The base compositions of ITS1 sequences of 5 tilapias species had little difference. From the UPGMA cluster resuh, the O. niloticus and O. niloticus × O. aureus was in the same branch, O. mossambicus and O. aureus was in the same branch. After O. mossambicus and O. aureus clustered, O. mossambicus × O. niloticus clustered with them. [ Conclusion ] Phylogenetic analysis indicated that the five species can be divided into three groups, among which the group of niloticus and Ni-ao tilapias is closely , aureus and mossambieus is closely , red tilapias is closely with aureus- Mozambique group.
