摘要
[目的]为湖北白鹅肉质性状的间接选择提供参考依据。[方法]采用聚丙烯酰胺凝胶电泳(PAGE)检测湖北白鹅基因组DNA 8个微卫星标记多态性,计算基因频率(P)、平均基因杂合度(H)、多态信息含量(PIC),分析白鹅性别、日龄及基因型与肌肉肌苷酸含量的相关性。[结果]所检测的8个微卫星座位的平均等位基因数为2.75个,H为0.594 5,PIC为0.508 7。56日龄鹅肉样肌苷酸含量为(1 360.25±82.56)mg/g,显著高于180日龄鹅肉样肌苷酸含量(1 088.07±63.56)mg/g。所检测的8个微卫星座位,ADL166座位AA基因型、ADL210座位AA与BC基因型、MCW0104座位AB与BC基因型、MCW264座位AA基因型、MCW0085座位AA与BB基因型、MCW0004基因座BC基因型、MCW0120座位AC基因型、MCW0014座位AA基因型个体的肌肉肌苷酸含量较高。[结论]这些微卫星标记有望作为遗传标记对鹅的肉质性状进行标记辅助选择。
[ Objective ] The reference for the indirect selection of Hubei white goose meat quality was provided through the analysis. [ Method] The polymorphisms of 8 micro-satellite markers of the Hubei white goose population were detected with the polyacrylamide gel electrophoresis(PAGE). The rate of alleles, heterozygosity and polymorphism information content (PIC) were calculated and the relationship between sex, age, genotype of goose and the content of inosine-5-monophosphate(IMP) in muscle was analyzed. [ Results] The results showed that in Hubei white goose population, the number of alleles was 2.75 ; the mean heterozygosity(H), 0. 594 5 and PIC, 0. 508 7 for the 8 mierosatellite loeuses. The content of IMP in muscle of geese at age of 56 d was (1 360.25 ±82.56) mg/g,whieh was higher significandy than that ( 1 088.07 ± 63.56) mg/g of geese at age of 180 d. For the 8 micro-satellite locuses, the higher content of IMP in muscle of geese appeared in the following genotype: the genotype of ADL166 locus was AA; ADL210 locus, AA and BC; MCW0104 locus, AB and BC; MCW264 locus, AA; MCW0085 locus, AA and BB; MCW0004 locus, BC; MCW0120 locus, AC and MCW0014 loeus, AA. [Conclusion] The microsatellite markers could be used as the genetic market for the improvement of Hubei white goose meat quality in breeding and selection.
