BCP荧光染色法诊断疟疾的实验研究 被引量:5

摘要 采用荧光染色剂苯并硫羟基嘌呤(benzothiocarboxypurine,BCP)对疟原虫进行染色实验,同时用姬姆萨染色做对照。BCP染色后疟原虫厚血膜在400倍荧光显微镜下见,疟原虫各发育期及滋养体、裂殖体和配子体呈特异性苹果绿色荧光;胞桨疟色素呈黄色;白细胞胞桨染成亮绿色。血膜中的异物不呈荧光色,易区别。储存120d后的厚血膜着色效果亦较好。BCP染色恶性疟与间日疟血片,敏感性为97.4%、91.2%,特异性为66.7%、50%,阳性预测值97.6%、88.6%阴性预测值50%与40%。认为BCP染色法是一种有使用价值的疟疾诊断新方法。 The fluorochrome benzothiocarboxypurine (BCP) was used for plasmodium staining in malaria diagnosis,with Giemsa as control group.The results showed that parasite including trophozoite,schizont and gametocyte chromation fluoresced bright applegreen and the cytoplasm yellow under the fluoro microscopy (x400).Leucocytes stained bright green.There was no visible interference by non parasitic material,which was easily distinguished.The film stored for 120 days was stained good.It indicated that BCP method had a sensitivity of 974%,a specifity of 66.7%,a positive predict value (PV+) of 976% and a negative predict value (PV-) of 50% for Plasmodium falciparum.For P.vivax it were 91.2% sensitivity,50% specificity,88.6% PV+ and 40 PV-, respectively.It revealed that BCP fluorochrome stain is a valuable method for malaria diagnosis.
