摘要
以达维、金铃、玉坠、平顶黄、薄壳红等5个平欧杂交榛品种茎段和根蘖苗茎段为外植体,进行适宜基本培养基的筛选、不定芽的诱导和快速繁殖研究。结果表明:(1)以茎段和根蘖苗茎段为外植体进行初代培养,污染率分别为75%以上和20%以下,外植体存活率分别为15%以下和65%以上,增殖倍数分别低于0.3和高于1.5;(2)在NRM、DKW、WPM、NN、MS等5种基本培养基中,NRM培养基最适宜平欧杂交榛品种试管苗的生长,其茎长和平均芽数高于其它培养基;(3)培养基NRM+5mg·L-16-BA+0.01mg·L-1IBA+32.21mg·L-1腐胺+29.05mg·L-1亚精胺+10.10mg·L-1精胺有利于外植体芽体的萌发与生长;(4)试管苗蘸1g.L-1IBA20s进行试管外扦插生根,15d后生根率可达95%以上,移栽成活率在97%以上。
Stem segment and root tiller explants of hybrid hazelnut cultivar Dawei,Jinling,Yuzhui,Pingdinghuang and Bokehong were used to select proper basic medium,establish buds inducement and rapid propagation procedure.The results showed that:(1)During primary culture process,stem segment and root tiller explants were cultured in DKW medium,and the contaminate rate was above 75% and below 20%,survive rate of explants were below 15% and above 65%,propagation times were below 0.3 and above 1.5 respectively;(2)Among basic medium NRM,DKW,WPM,NN and MS,shoot length and auxiliary buds per shoot in NRM medium were higher apparently compared with other medium;(3)Medium NRM+5 mg·L^-1 6-BA+0.01 mg·L^-1 IBA+32.21 mg·L^-1 putrescine+29.05 mg·L^-1 spermidine + 10.10 mg·L^-1 spermine were profitable to promote buds germination and growth;(4)Tube plantlet could root in 15 d and rooting rate reached over 95% when its end dipped in 1 g·L^-1 IBA for 20 seconds,and its roots were strong.Transplant survive rate was over 97%.
