摘要
本实验应用降落PCR方法从聚球藻Synechococcus cedrorum中扩增出717bp的hox1基因全序列,并与从Gen-Bank下载13个蓝藻hox1基因序列比较,用MEGA4.0进行序列同源性分析及蛋白结构特性分析。结果表明,Synecho-coccus cedrorum的hox1基因与聚球藻Synechococcus PCC7942和钝顶节旋藻Arthrospira platensis的hox1基因序列同源性分别为99.9%和99.2%,氨基酸序列同源性分别为99.6%和97.9%。还用邻接法和最大简约法构建了系统树并进行了聚类分析。
The complete hox1 gene including 717bp was cloned from Synechococcus cedrorum by using touchdown PCR technology.Homology analysis based on cyanobacteria hox1 gene showed that the similarity is 99.9% between Synechococcus cedrorum and Synechococcus PCC7942 and 99.2% between Synechococcus cedrorum and Arthrospira platensis.The similarity of deduced amino acid is 99.6%(between Synechococcus cedrorum and Synechococcus PCC7942) and 97.9%(between Synechococcus cedrorum and Arthrospira platensis) respectively.The phylogenetic relationships among eight cyanobacteria were constructed by the methods of neighbor-joining(NJ) and maximum parsimony(MP).
