摘要
目的:探讨细胞分化诱导剂全反式维甲酸对人体肝癌细胞几种酶活性的影响。方法:用 10μmol/L全反式维甲酸处理 BEL-7402人肝癌细胞,观察细胞碱性磷酸酶(ALP)、酪氨酸-α-酮成二酸转氨酶(TAT)和γ-谷氨酸转肽酶(γ-GT)活性以及甲胎蛋白(AF P)分泌量。结果:10μmol/L全反式维甲酸使反映肝细胞恶变的指标 AFP分泌量和 γ-GT比活力明显下降( P< 0.05或<0.01),ALP比活力和反映肝细胞分化的TAT比活力则明显升高(P<0.01或<0.05)。这些酶学指标与AFP分泌量的变化有助于综合反映肝癌细胞的再分化。
Objective: To study the effect of all trans-retinoic acid (ATRA)on the activities of several enzymes of human hepatocarcinoma cells. Methods: The BEL-7402 human hepatocarcinoma cells were treated with ATRA, and the activities of alkaline phosphatase (ALP ), tyrosine-α- ketoglutarate transferase(TAT), γ-glutamyl transpeptidase (γ-GT), and the secretory amount of alpha-fetoprotein (AFP) of the cells, were determined. Results: Under the action of ATRA in the concentration of 10 umol/L, the secretory amount of AFP and the specific activity of γ-GT, which reflect the canceration of hepatocytes , decreased (P <0.05 and P <0.01, respectively ); whereas the specific activities of ALP and TAT, which indicate the differentiation of hepatocarcinoma cells, increased (P < 0. 01 and P < 0. 05, respectively). Conclusion: The observation of these enzymological indexes and the secretory amount of AFP may be a synthetical reflection of the redifferentiation of hepatocarcinoma cells.
