摘要
鱼类生长素的纯化现一般采用三步法,即脑垂体碱性提取物经凝胶过滤柱(Sephadex),离子交换(DH-52),最后经反相柱(ODS-120T)而纯化.如第一步提取后生长素纯度高,可以省去中间离子交换层析这一步,但本研究表明,即使生长素含量不高,通过仔细切会同一峰内的杂蛋白,仅收集GH含量高的级分.采用二步法也可得到良好的分离.各种二步法均试用过,仅有反相层析给出最好的分辨力,分离的草鱼生长素经分子量测定、氨基酸组成、反相色谱等色谱行为证明是单一的组分.
With two step column chromatography,growth hormone from grass carp are isolated.SDSPAGE.chromatography behaviours,and amino acid composition show that they are homogenious growth hormone.Generially,the isolation of growth hormones from fish need three column:Sephadex,DE-52 and ODS-120T,but in this report,by carefully collect the GH-rich sample,the column DE-52 is omitted,the GH is purified as well.
