摘要
目的:探讨表皮生长因子(EGF)、碱性成纤维细胞生长因子(bFGF)、胰岛素样生长因子-1(IGF-1)、L-谷氨酰胺(Gln)、精氨酸、重组人生长激素(rhGH)和胰岛素对原代培养的小肠粘膜上皮细胞分化的影响。方法:孕龄17~19天Wistar系孕鼠胎仔小肠制备成小肠绒毛细胞团悬液,培养48h后加入上述肠营养素,通过测定ALP活性观察不同剂量肠营养素对贴壁生长的肠上皮细胞分化的作用。结果:当肠营养素刺激肠上皮细胞增殖时,就能直接影响细胞分化。EGF、IGF-1、Gln、胰岛素、rhGH促进肠上皮细胞分化,而精氨酸、bFGF则可使细胞分化降低。结论:肠营养素不仅有营养的作用。
Objectives:To investigate the effects of EGF,bFGF,IGF 1,Gln,arginine,rhGH,and insulin on the differentiation of small intestinal epithelium in the primary culture. Methods:The epithelial organoids of small intestine were obtained from wistar fetal rats,and were successfully cultuerd.After 48h incubation,each group received one kind of intestinal nutrients at different levels alone.Alkaline phosphatase(ALP) activity of epithelial cells was measured to identify the differentiation of small intestinal epithelium. Results:While it had trophic effect,the intestinal nutrient could exert direct influence over the epithelial differentiation of small intestine.EFG,IGF 1,Gln,insulin,and rhGH accelerated cell differentiation,but arginine and bFGF decreased epithelial differentiation. Conclusions:Besides their trophic effects,the ability of intestinal nutrients to induce the differentiation of intestinal epithelial cells must be paid more attention.
