不同牙科烤瓷合金材料诱导小鼠成纤维细胞DNA损伤的研究 被引量:5

摘要 研究不同牙科烤瓷合金材料诱导小鼠成纤维细胞(L929细胞)DNA的损伤情况,评价各种合金材料的遗传毒性。选择镍铬合金、钛合金、钴铬合金和金合金4种牙科烤瓷合金材料,制备4种合金材料的浸提液,用浸提液培养L929细胞。采用单细胞凝胶电泳(彗星试验)方法检测细胞DNA的损伤情况,镍铬合金和钛合金对细胞DNA损伤等级主要为重度损伤,钴铬合金组和金合金组的DNA损伤等级分别为中度损伤和轻度损伤。4种合金组彗星细胞的尾长、尾矩和Olive尾矩数值之间的差异具有统计学意义(P<0.05)。不同牙烤瓷合金材料诱导L929细胞DNA损伤的情况表现不同。金合金的遗传毒性性能优于镍铬合金、钛合金和钴铬合金,是具有良好生物学性能的牙科烤瓷合金材料。 The aims of this study was to investigate the DNA damage of mouse fibroblast(L929 cells) induced by different kinds of dental ceramic alloy and to evaluate the genotoxicity of alloys. Extractions of four kinds of alloy (Ni- Cr alloy, Ti alloy, Co-Cr alloy and Au alloy) were prepared and used to culture L929 cells. DNA damage of L929 cells were detected by the method of single cell gel electrophoresis (SCGE). The ranks of DNA damage of Ni-Cr alloy group and Ti alloy group were severe damaged, and that of Co-Cr ally group and Au ally group were damaged in midrange and slight damaged, respectively. The values of three comet parameters, tail length, tail moment and Olive tail moment, were significantly different among four kinds of dental ceramic alloy. The DNA damage (of L929 cells induced by four kinds of dental ceramic alloy was different. The genotoxicity of Au alloy was superior to that of Ni-Cr alloy, Ti alloy and Co-Cr alloy. Au aloy was a kind of dental ceramic alloy with good biocompatibility.
