蛋白酶激活受体2在断奶腹泻仔猪胃肠道组织中的定位及其作用 被引量:1

摘要 以体质量均约为20 kg的10头断奶仔猪为研究对象(腹泻仔猪7头、健康仔猪3头),采用常规HE染色法观察腹泻仔猪胃肠道病理组织的变化,采用免疫组化检测蛋白酶激活受体2(Proteinase-activated receptar-2,PAR-2)的量。结果显示,通过HE染色观察发现病理组织学变化主要表现为粘膜上皮细胞脱落变性,肠壁平滑肌变薄,固有层内中性粒细胞、嗜酸粒细胞、淋巴细胞等浸润、水肿,黏膜下层淋巴小结增生,分别表现出不同程度的炎症。运用免疫组织化学分析发现仔猪胃肠道广泛的表达PAR-2,特别在黏膜上皮细胞的顶端、腺细胞的基底部、血管上皮细胞、成纤维细胞、中性粒细胞、嗜酸性细胞等,所设对照组均未见阳性反应。并且,PAR-2在腹泻仔猪比正常仔猪的表达增加,特别是炎性细胞有强表达。以上结果表明,PAR-2在仔猪腹泻的病理过程中具有一定的作用。 The investigative object were ten piglets (7 diarrhea piglets, 3 normal piglets) whose average weight were about 20 kg. HE staining was used to survey pathological changes in GI tracts of diarrhea piglets,and immuno- histochemistry was used to detect quantity of PAR-2. The results showed that most pathological changes showed desquamation and denaturation in the epithelium mucosae, smooth muscle thinning of bowels, infiltrated neutrophil and eosinophil in the lamina propria with HE staining, and lymph nodules were hyperplasia in submucosa. They showed different infammation degrees. Immunohistochemical analyses clearly showed abundant espression of PAR-2 in the GI tract of piglet,especially in the apical region of epithelium mucosae and basular part of gland cell,and was found in vascular endothelial cell, fibroblast, neutrophlie granulocyte, eosinophil cell and so on. But control group was found no positive reaction. Further, immunoreaction of PAR-2 was more expressed in the diarrhea pigs than in the normal ones,especially in the inflammatory cell. The above results indicated that PAR-2 did determinate use in diar-rheal process of piglet.
