朝鲜族DXS6797STR位点的遗传多态性分析

摘要 [目的]研究延边地区朝鲜族人群短串联重复序列(STR)基因座DXS 6797的遗传多态性,获得该基因位点的群体遗传学数据.[方法]应用PCR扩增技术及聚丙烯酰胺凝胶电泳方法,对106名朝鲜族女性个体和186名朝鲜族男性个体的DXS 6797 STR基因座等位基因频率进行分析.[结果]在女性样本DXS 6797基因座中各检出7种等位基因和16种不同的基因型,分析基因型观察值和期望值,基因座多态性分布符合Hardy-Weinberg平衡定律,杂合度为0.717,多态信息含量为0.68,个人识别率为0.876;在男性样本DXS 6797基因座中共检出7种等位基因,单倍型多样性为0.745 5.[结论]STR基因座DXS 6797具有较高的个人识别力,可应用于法医学检验和朝鲜族人群遗传学分析. OBJECTIVE To understand the allele structure and genetic polymorphism at DXS 6797 short tandem repeats (STRs) locus in Korean-Chinese nationality, Yanbian, and to establish the related genetic database. METHODS The allele frequencies of the DXS 6797 locus in 106 unrelated female individuals and 186 unrelated male individuals from Korean-Chinese nationality were analyzed by polymerase chain reaction (PCR) and polyacrylamide gel electrophoresis (PAGE). RESULTS Seven alleles and sixteen genotypes were observed at DXS 6797 locus in the female individuals, respectively, and met Hardy-Weinberg equilibrium. The statistical analysis of DXS 6797 STR locus showed the heterozygosity was 0. 717, the polymorphic information contents (PIC) was 0.68; the power of discrimination (PD) was 0. 876; and also seven alleles were observed at DXS 6797 locus of the male individuals, the haplotype diversity reached 0. 745 5. CONCLUSION DXS 6797 STR locus has high heterozygosity, so it can be applied to forensic medicine and Korean-Chinese nationality.
