摘要
根据已克隆的野桑蚕酚氧化酶原基因的cDNA序列,利用生物信息学工具对该基因编码蛋白质的部分性质进行了分析.结果表明:野桑蚕酚氧化酶原基因PPO1和PPO2编码蛋白质的分子量和等电点分别为78.703 kDa,80.075 kDa和6.28,5.62;氨基酸序列的C-末端均没有疏水跨膜区域,其基因编码蛋白质的三级结构分别具有12个α螺旋、16个β折叠和14个α螺旋、18个β折叠.对18个物种的基因进化进一步分析表明:野桑蚕与家蚕的亲缘关系最近,与其他物种的亲缘关系相对较远.
The partial characterization, based on the cloned cDNA sequence of PPO gene from Bombyx mandarina and used bioinformatic tools, was analyzed. The results revealed that PPO1 and PPO2 gene of Bombyx mandarina encode the proteins, with a predicted molecular weight of 78. 703 kDa,80. 075 kDa and isoelectric point of 6.28,5.62, respectively, no hydrophobic membrane- spanning regions at its C- terminus of amino acids sequences, and 12 α - helix, 16 β - sheet, 14 α- helix,18 β- sheet in its tertiary structure, respectively. The phylogeny of PPO genes was further analyzed from 18 different most closely related to Bombyx mori, but far from other species.
