摘要 目的:观察Tween80联合42℃温热对人卵巢癌SKOV-3细胞热耐受和热休克蛋白(heat shock protein,HSP70)表达的影响。方法:采用MTT法测定Tween80联合42℃温热作用后,SKOV-3细胞的生长抑制率改变;采用免疫细胞化学法测定Tween80联合42℃温热作用后SKOV-3细胞的HSP70表达情况。结果:Tween80联合42℃温热组的SKOV-3细胞生长抑制率较单纯作用组明显升高(P<0.05);Tween80联合42℃重复温热组的细胞增殖率较单纯42℃重复温热组明显下降(P<0.05);不同体积分数(0.025%、0.05%、0.075%和0.1%)的Tween80联合42℃温热组与单纯温热组相比,SKOV-3细胞的HSP70表达均下降,并且随着Tween80浓度增加,HSP70的表达逐渐下降。结论:Tween80联合42℃温热对人卵巢癌SKOV-3细胞的生长抑制可能有相加或协同作用,Tween80可抑制加热后细胞的HSP70表达和热耐受性产生。 Objective:To investigate the effects of Tween 80 combined with 42℃ hyperthermia on the thermotolerance and the expression of heat shock protein (HSP70) in the human ovarian cancer cell line SKOV-3. Methods:The inhibition ratio of cell proliferation was detected by MTT assay after treatment with Tween 80 combined with 42℃ hyperthermia. The expression of HSP70 was detected by immunocytochemical method. Results : The inhibition ratio of proliferation of SKOV-3 ceils was greatly increased and the proliferation ratio was obviously decreased after the combined treatment with Tween 80 and 42℃ hyperthermia compared with that of the Tween 80 or 42℃ hyperthermia single treatment group (both P 〈 0.05 ). The expression of HSP70 protein was decreased after combined treatment with different concentrations of Tween 80 (0. 025%, 0.05% , 0. 075% , and 0.1% ) plus 42℃ hyperthermia compared with the groups treated with single 42℃ hyperthermia. With the increase in the concentrations of Tween 80, the HSP70 expression was decreased gradually. Conelusion:Tween 80 combined with 42℃ hyperthermia has additive or synergistic effect in inhibiting the proliferation of SKOV-3 cells. Tween 80 can inhibit the expression of HSP70 and thermotolerance of the ceils after hyperthermia.
