摘要
报道由我国云南省土壤中分离的一株放线菌所产生的广谱抗病毒抗生素17997[1]的细胞毒性及作用机制研究。用氚标记物参入法测定抗生素17997对VERO细胞DNA、RNA及蛋白质合成的TD50(50%毒性剂量)分别为76.8±2.2、130.9±19.5及144.2±17.1μmol/L。抗生素17997无毒浓度不影响单疱病毒Ⅰ型的吸附及释放;对病毒无直接灭活作用;感染后6h给药无抑制病毒作用;对病毒逆转录酶及DNA聚合酶均无抑制作用。抗生素17997可能作用于病毒复制早期阶段。
Antibiotic 17997 is a broad spectrum antiviral antibiotic produced by a strain of Streptomyces isolated from a soil sample collected in Yunnan Province, China. It shows strong inhibitory effects against Herpes virus and Retrovirus both in vitro and in vivo. The toxicities of antibiotic 17997 on VERO cells have been estimated by the method of incorporation of tritium labeled compounds. The TD50for the synthesis of DNA, RNA and protein are 76.8±2.2, 130.9±19.5 and 144.2±17.1μmol/L respectively. The nontoxic concentration of antibiotic 17997 did not directly inactivate HSV1 infectivity; did not affect the adsorption and release of HSV1 from VERO cells; did not inhibit HSV1 replication when treatment was strated before infection or after 6h infection; did not show inhibitory effects on HIV1 reverse transtcriptase and DHBV replication comples DNA polymerase. These results indicate that antibiotic 17997 might affect the early stage of HSV1 replication.
