抗人Ig抗体诱导人乳腺癌MCF-7细胞凋亡的实验研究

摘要 目的观察抗人Ig抗体对人乳腺癌传代细胞系MCF-7细胞的凋亡诱导作用。方法用培养上清中加入抗体和原位电转的方法将抗人Ig抗体分别作MCF-7细胞的抗体封闭实验,用流式分析仪进行细胞凋亡检测。结果培养上清中加入抗人IgG、抗IgM抗体后,对MCF-7细胞系没有明显的凋亡诱导作用。而通过原位电转的方法将抗人IgG、抗人IgM抗体转入MCF-7细胞内,发现二者均对MCF-7有明显的促凋亡作用(P<0.01)。结论MCF-7细胞内的IgG及IgM对MCF-7细胞的生存具有重要意义。 Objective To observe blocking effects of anti-human Ig antibodies on apoptosis of human breast cancer cell line MCF-7. Methods we observed the apoptosis of human breast cancer cell line MCF-7 by adding anti-human Ig antibodies to culture medium or by transporting them into MCF-7 cells, we detected the cell apoptosis by flow cytometry. Results There is no appreciable difference on apoptosis can be observed when adding anti-human lgG and IgM to culture supernatant of MCF-7 cell, Whereas we observed the apoptosis of MCF-7 significantly increasing after transporting the anti-human IgG or IgM antibody into it by in situ electroporation( P 〈 0.01 ). Conclusion These data conform that blocking IgG ,IgM that in cytoplasm of MCF-7 can induced the apoptosis of these cells.
