免疫多糖对养殖南美白对虾作用的研究 被引量:152

THE EFFECT OF IMMUNOPOLYSACCHARIDE AS A FOOD ADDITIVE ON THE PENAEID SHRIMP, PENAEUS VANNAMEI

摘要 于1995年6月在威海市水产开发公司对虾育苗场采集南美白对虾,经暂养后,选取体长6-7cm的个体,利用由海藻中提取的与微生物多糖有类似结构和性质的免疫多糖作为饵料添加剂,以口服形式对南美白对虾进行免疫,刺激其免疫系统。通过连续测定南美白对虾血清中酚氧化酶活力和肌肉组织液中的抗菌、溶菌活力,以及超氧化物歧化酶活力,研究免疫多糖对南美白对虾免疫功能、机体抗氧化能力的作用。结果表明,除个别情况外,实验组南美白对虾的酚氧化酶活力,溶菌、抗菌活力和超氧化物歧化酶活力,均高于对照组的。由此推断,免疫多糖具有增强南美白对虾免疫功能的作用,并有可能促进其机体的抗氧化能力。 Immunopolysaccharide (IPS), a product exbeted from sea algae, with the characterishcssimilar to those of microbial polysascharides, was used as a food addihve to test itS effechveness ofantidisease. Aroun 100 juvehile Penaeus vannamei with a body length of 3-4cm were collectedfrom the Shrimp Hatchery of Weihai Aquaculbe Company on Jun 2, 1995, and were then nursedin 4 PVC wh (100cm ×150cm ×100cm) in the Aquarium of the Institute of Oceanology, TheChinese ACademy of Sciences. Water tempethe was kept at 22-28t, salinity 32. The warer wasaerated, and chaned tWice a day (1 / 3-1 / 2 volurn of water was renewed each hme). No IPSwas addd in the food during nmeery periods.After nmeery, Pvannamei with a bod length of 6-7cm were shifted to two PVC tanks of thesame size (diametCr 90cm, height 100cm, one for control, the othr for test grouP, stocking density18 pieces / tank on August 18, 1995, when experiments of superoxide dismutase (SOD) activity,bacteriolytic (UL) and antibacterial (Ua ) activities began. The controlled group was fed with foodwithout IPS, while the test group was fed with IPS. The maintenane of shrimps was the same asthe nursery period.Experiment of phenoloxidase (PO) test was designed as the same as those of SOD UL and Uatest excePt that for the stocking density of 16 pieces / tank, only tWo consecutive samples weretaken because of insufficent number of sdrimp. Shrimp samples with a body length of 6-8cm werecollected from october 1, 1995.Blood samples were collected from the hearts of 4-5 shrimps to measure PO activities for Pvannamei. Two consecutive blood samples (each sample consisted of contrlled and test groups) werecollected in an 20 day's interval and the blood serum was prepared at 4t to measure their POactivities using a Spectrometerr751. Muscle samples were chn from 2-5 shrimps in 10 day'sintervals to measure SOD activity, UL activity to Micrococcus lysoleikticus and Ua activity toEscherichia coli of P vannamei Three consecutive muscle sampes (each consisted of controlled andtest groups) were collected and the supertant of muscle homogenare was prepared by centrifugation at4℃ to measare their PO, UL and Ua astivities using the Spectrometer-751.The results show that PO, SOD UL and most Ua activities of P. vannamei of the tost groups aregenerally higher tnan those of the controlled groups, indicating the effechveness of IPS in enhancingPO, UL, Ua and SOD activities of P vannamei, leading to an increase in the immunological andanti - oxidation activities (Tab. 1 and Tab.2). Furthr studies are needed to identify the mechedsm ofIPS in antidisease effect and the relationships among SOD PO, UL and Ua activities.
