实验性脑损伤后海马、血浆内皮素含量变化及意义

摘要 目的研究ET-1对脑损伤后海马神经原坏死的影响。方法Wistar大鼠50只分为对照组、4组脑损伤,脑损伤采用Feeney等的落体撞击法造成右顶叶局限性脑损伤,分别于伤后30分钟、6、24及72小时处死取材;用RIA法测定血浆、海马匀浆中ET-1含量变化。结果血浆及海马ET-1水平在脑损伤后各时间点均明显高于对照组,且ET-1含量变化与病程有关。结论脑损伤后ET-1增高是引起海马缺血、神经原坏死的重要原因;动态测定血浆ET-1水平可作为脑损伤后病情发生、发展评价指标。 bjective In order to study the effects of ET-1 on causing of hippocampal neuron after brain trauma. Methods Fifty Wistar rats were randomly divided into five groups.Normal contrast group and four brain trauma.The lesion of localized right parietal lobe was made by Feeney′s methods.The levels of ET-1 in the plasma and hippocampus of rat with brain trauma were detered by RIA after lesion 30min;6h;24h and 72h. Results Levels of ET-1 in the plasma and hippocampus of rat with brain trauma were significantly higher than control group and changes of ET-1 were correlated with the course of disease. Conclusion These suggest that increasing of ET-1 after brain injury may be an important reason causing ischemia and death of hippocampal neuron;and dynamic observation of ET-1 in the plasma can be used is an criteria of occurrence and development of brain injury
