丙戊酸钠对Kasumi-1细胞VEGF、bFGF表达的影响

摘要 目的探讨丙戊酸钠(VPA)对白血病细胞株Kasumi-1细胞生长及血管新生的作用机制。方法不同浓度VPA处理Kasumi-1细胞不同时间,RT-PCR及Western Blot法检测Kasumi-1细胞血管内皮细胞生长因子(VEGF)、碱性成纤维细胞生长因子(bFGF)mRNA及蛋白水平。结果VPA作用后VEGF mRNA及蛋白表达下调,bFGF mRNA表达下调,均呈剂量依赖性。结论VPA可通过下调VEGF及bFGF表达,抑制血管新生,进而抑制白血病细胞增殖。 Objective To investigate the mechanism of action of valpreie acid (VPA) on the proliferation and inhibition of angiogenesis of Kasumi-1 cell. Methods Kasumi-1 cell was treated with VPA in different concentrations for different time. The mRNA and protein expression of VEGF and bFGF were detected by RT-PCR and Western Blot assays. Results After treated by the VPA, the mRNA and protein expression of VEGF decreased , the expression of bFGF mRNA increased, both in dose-dependent manner. Conclusion VPA can inhibit the angiogenesis and proliferation of Kasumi-1 cell, by regulating the expression of VEGF and bFGF.
