摘要
用光学显微镜和透射电子显微镜对胭脂鱼精子的结构进行研究,并在光学显微镜下对精子各部分的长度进行测定。结果表明,胭脂鱼的精子主要分为头部、中段和尾部(鞭毛);头部在光学显微镜下近圆形,透射电镜下头部纵切似马蹄形;头部无顶体,主要由细胞核组成,核内染色质致密,核内有核空泡;核外可见清晰的核膜,头部几乎无细胞质存在;头部后端中央处有一较深的植入窝,深度占细胞核总长约2/3,植入窝内有中心粒复合体。精子的中片紧连在核的后端,由中心粒复合体和袖套组成;袖套两边接近对称;中心粒复合体由近端中心粒和远端中心粒组成;近端中心粒与远端中心粒之间的角度近直角,呈"T"字形;精子尾部轴丝与细胞膜之间有较大的空间,轴丝与细胞膜之间还有囊泡等细胞器;精子尾部有侧鳍;尾部可分为主段和末段,尾部主段具有典型的"9+2"的轴丝结构。光学显微镜测定结果表明,精子头长约(2.56±0.14)μm,尾部约(52.69±5.59)μm,全长约(55.51±5.67)μm。
Structure of Myxocyprinus asiaticus (Bleeker) spermatozoon was studied with light microscope (LM) and transmission electron microscope (TEM). The lengths of different parts of 31 spermatozoa were measured under LM, and the results showed that the spermatozoon ofMyxocyprinus asiaticus could be mainly divided into three parts :head, midpiece and tail (flagellum). Head of spermatozoon was spherical shaped under LM and horseshoe shaped under TEM when it was longitudinally sectioned, which mainly consisted of nucleus, without acrosome. The chromatin in nucleus was dense, nuclear vesicle could be seen in the nucleus, and nuclear membrane could clearly be seen outside of the nucleus. Only a little cytoplasm was existed in head. There was a deep implantation fossa at the caudal-contral end of the nucleus, the depth of which was about 2/3 of the length of head,with a centriolar complex. Midpiece was at the posterior end of the nucleus, and consisted of almost symmetrical sleeves and centriolar complex, which composed of proximal centriole (PC) and distal centriole (DC). The angle of PC and DC was almost right,ie. "T" shape like. A big space existed between the axial filament and cell membrane, and there were vesicles and cytoplasm in the big space. The tail could be divided into two parts (principal piece and end piece), and with lateral fins on both sides of the flagellum. The principal piece of the spermatozoon tail was long and possessed a conventional "9 + 2" structure. The light microscopy measurement showed that head length,tail length and total length were about (2.56±0. 14)μm, (52. 69±5.59)μm and (55.51 ±5.67) μm respectively.
