siRNA抑制Wnt-1基因对人肺腺癌细胞株A2的影响

摘要 目的:研究siRNA对人肺腺癌细胞株A2中Wnt-1基因表达对细胞生物学行为的影响。方法:将siRNA用脂质体转染A2细胞,RT-PCR测定Wnt-1 mRNA表达变化;Western blotting测定Wnt-1蛋白表达的变化;流式细胞仪检测对细胞凋亡的影响。结果:与对照组相比,传染siRNA后,实验组靶基因mRNA表达和蛋白表达均有明显降低,细胞凋亡增加。结论:Wnt-1特异性siRNA能有效干扰A2细胞内其相应基因的表达,而靶基因表达受到抑制后细胞凋亡增加。 Objective:To evaluate the effectiveness of siRNA on Wnt-1 gene expression in human lung adenocarcinoma cell line A2 ,so as to asses the role of Wnt 1 in A2 cells. Methods :SiRNA targeting Wnt-1 mRNA were transfected into A2 via Lipofectamine. The changes of expression of Wnt 1 mRNA were assayed by RT PCR. Using Westen blot ting method to detect the changes of expression of Wnt 1 protein. MTT was used to detect the inhibition of cells growth. Flowcytometry method was used to detect the apoptosis. Results :Compared with control group, the expression of mRNA and protein of Wnt 1 in A2 cells were obviously decreased in siRNA transfected ceils. The cell growth and viability of siRNA transfeeted group were significantly inhibited(P〈0.05). The apoptosis of A2 cells increased too after siRNA transfection. Conclusion:Interference with siRNA against Wnt-1 can effectively inhibit the specific gene expression, which may play a significant role in the inhibition the proliferation and the apoptosis of the A2 cells.
