摘要
目的:观察不同移植时间窗对静脉移植骨髓间充质干细胞(MSCs)在大鼠损伤脊髓内存活和迁移的影响,探讨移植的最佳时间。方法:体外分离、培养、纯化MSCs,改良Allen法制造大鼠脊髓损伤(SCI)模型,40只SD大鼠随机分为假手术组、SCI后0、6、12、24h,3、5和7d移植组,移植组经尾静脉注射Hoechst33342标记的MSCs,移植后7d取脊髓组织,观察各组MSCs在脊髓损伤处的存活和迁移情况。结果:SCI后0、6、12、24h移植4组,脊髓损伤处的MSCs的存活数量较少,但迁移距离较长;SCI后3d移植组,脊髓损伤处MSCs的存活数量较多,且迁移距离较长;SCI后5和7d移植2组,脊髓损伤处的MSCs的存活数量较少,且迁移距离较短。结论:在SCI后3d是静脉移植MSCs治疗大鼠SCI的最佳移植时间窗。
Objective: To study the influence of different transplantating time windows on the survival and immigration of marrow mesenchymal stem cells (MSCs) in injured spinal cord by intravenous administration, and to evaluate the most optimal intravenous administration time for MSCs. Methods: MSCs from adult rats were cultured. The rat spinal cord injury (SCI) model was prepared according to the modified Allen method. 40 SD rats were randomly divided into a non-injured group,and seven transplanted groups at 0 h, 6 h, 12 h, 24 h, 3 d, 5 d and 7 d after SCI. MSCs labeled with Hoechst33342 were transplantated by tail vein. The survival number and migrating distance of MSCs in the spinal cord of all groups were measured. Results: The survival number of MSCs was fewer, but the immigration distance of MSCs was longer at 0 h, 6 h, 12 h and 24 h after SCI. The survival number of MSCs was more,and the immigration distance of MSCs was longer at 3 d after SCL The survival number of MSCs was fewer, and the immigration distance of MSCs was shorter at 5 d and 7 d after SCI. Conclusion: Three days after SCI is the most optimal transplantating time window by intravenous administration for MSCs.
