摘要
将含有gusA基因的质粒pAct1-D包裹在钨粉上轰击籼稻初生愈伤组织,通过检测gusA基因在籼稻细胞中的瞬间表达情况,对影响基因枪法转化效率的因素进行了研究.这些因素包括微粒加速装置参数、微粒参数、生物参数及其它参数.实验表明,在合适的转化条件下,籼稻品种青风占12、穗优占的最高转化效率分别为439。
Primary calli of indica rice were bombarded with tungsten particles coated with the plasmid pAct1D containing gusA gene. Factors influencing biolistic transformation efficiency were studied by detecting transient expression of gusA gene in indica rice cells. The factors included particle accelerator parameters, microprojectile parameters, biological parameters and other parameters. Under appropriate transformation condition, the highest transformation efficiency achieved is 439 and 915 blue spots per plate for indica rice variety Qingfengzhan 12 and Suiyouzhan, respectively.
