摘要 [目的]改进松辽黑猪脏器总RNA的高效快速提取方法,为检测脏器组织RNA提供基础。[方法]采用液氮研磨,利用Trizol使细胞膜破裂,并通过氯仿、异丙醇、无水乙醇处理,使RNA与杂质分离,得到高质量的RNA。[结果]得到的松辽黑猪脏器总RNA浓度和纯度都比较高,并且未被降解。[结论]该方法提取的RNA质量没有DNA污染、纯度高、没有降解、完整性好,稳定可靠,并且用时少。 [Objective] The aim was to improve an effective and rapid method for extracting total RNA of Songliao black pig, and provide basis for testing organs tissue RNA. [Method] Using liquid nitrogen grinding method, cell membrane was ruptured by using Trizol , through some treatments with chloroform, isopropanol and absolute alcohol, RNA was purified and high quality RNA was obtained. [Result] The concentration and purity were high of obtained total RNA of Songliao black pig, and were not degraded. [Conclusion] The quality and purity of RNA are high and not contaminated by DNA by using this method, which has better integrity and stability, and could save time.
