摘要
[目的]筛选1株生物表面活性剂高产量菌株,并对其发酵条件进行优化。[方法]利用血平板和油平板法,从油污水沟中分离得到1株生物表面活性剂高产菌,经形态、生理生化特征及16S rDNA鉴定,该菌为奇异变形杆菌E(Proteus mirabilisE)。对产生物表面活性剂的条件进行优化。[结果]结果表明,该菌在菜籽油15 ml/L(、NH4)2SO41.5 g/L、pH值为8、接种量6%,37℃、200 r/min发酵培养48 h,生物表面活性剂的产量达到4.1 g/L,是优化前的2.28倍。排油圈分析和TLC分析表明,菌株E发酵液排油圈直径为7.2 cm,表面活性剂为糖脂类生物表面活性剂;该生物表面活性剂对菜籽油乳化能力较好,可以使乳化性能稳定保持14 d以上。[结果]该研究结果为新型表面活性剂的开发和工业化生产奠定基础。
[ Objective] The purpose of this researeh was to fiher a high yield strain of biosurfactant and optimize its fermentation conditions. [Method] One biosurfactant-producing microorganism E was isolated from samples collected from polluted water with the methods of blood-plate and oil-plate. Based on the results of morphology, physiological and biochemical properties and phylogenetic analysis of 16S rDNA sequence, strain E was identified as Proteus mirabiliss E. The medium composition and the cultivation conditions were optimized by orthogonal and single factor experiment. [Result]The results indicated that the yield of biosurfactant under optimized conditions of 15 ml/L rapeseed oil, 1.5 g/L (NH4)2SO4, inoculation amount 6%, pH value 8 and 37℃ cultured 48 h at 200 r/min was 4.1 g/L, 2.28 times more than the preliminary preparation. TLC analysis of the ferment had classified the biosurfactant into glycolipid type. The diameter of oil displacement activity achieved 7.2 cm when use optimized medium. This biosurfactant had a good emulsification, the emulsified fluid could maintain at least 14 days. [collusion]This study will provide the basic reference for development and industrialization production of new biosurfactant.
