猪冠状动脉微栓塞模型血清TGF-β_1的表达 被引量:3

Expression of Transforming Growth Factor-beta 1 in the Model of Coronary Microembolization of in Pigs

摘要 目的:检测转化生长因子-β1(transforming growth factor-beta 1,TGF-β1)在猪冠状动脉微栓塞模型中的表达情况。方法:10只小型猪,通过经皮冠脉介入术于前降支注入微栓塞球建立冠状动脉微栓塞的模型,以ELISA法检测术前、术后2h、6h和1周时血清TGF-β1的浓度;RT-PCR法检测术后1周时心肌中TGF-β1 mRNA的表达情况。结果:与冠脉微栓塞术前(7.38±2.10ng·mL-1)相比,在术后2h血清中TGF-β1升高(14.86±4.04ng·mL-1),6h达到高峰(22.89±2.03ng·mL-1),1周时下降(20.44±3.99ng·mL-1),但仍高于术前;RT-PCR法检测示1周时前壁微梗死区心肌组织TGF-β1 mRNA水平表达高于后壁未梗死区。结论:冠状动脉微栓塞后血清TGF-β1呈升高趋势,且至少持续1周左右;1周时微栓塞相关心肌组织中TGF-β1 mRNA的表达也明显升高。 Objective:To detect the expression of transforming growth factor-beta 1(TGF-β1 ) in the model of coronary microembolization in pigs. Methods:The left anterior descending coronary artery of 10 pigs were embolized by injection of 42μm microspheres with a total number of 120,000. The serum level of TGF-β1 was detected by ELISA in different time point(base, 2 hours, 6 hours, 1 week). TGF-β1 mRNA from anterior and posterior myocardium were analyzed by RT-PCR at 1 week. Results:Compared with the base level of TGF-β1(7.377±2.10ng·mL^-1, TGF-β1 began to increase significantly at 2 hours(14.857±4.04 ng·mL^-1 after coronary microembolization,(P 〈0.05)Its peak level was 22.884±2.03ng·mL^-1at 6 hours. After 1 week, TGF-β1 began to decrese, which still higher than the base level. Results of RT-PCR demonstrated higher expression of TGF-β1 mRNA in the anterior myocardium compared with the posterior one. Conclusion:The serum level of TGF-β1 elevates after coronary microembolization, which remains at least one week. TGF-β1 mRNA also reach a higher expression in microemboliztion-associated myocardium at 1 week.
