鸡新城疫病毒内蒙古地方株F基因遗传变异分析

摘要 将3株新城疫病毒(NDV)内蒙分离株纯化培养后,提取病毒RNA,经RT-PER扩增F基因,并测定出F基因的全部核苷酸序列为1662bp,编码有554个氨基酸残基。与国内外发表的部分NDV的F基因核苷酸序列进行比较,结果表明这3株分离株核苷酸同源性分别为97.5%,97.9%和98.0%,与标准毒株F48E9的同源性分别为85.8%,86.4%和86.8%,与长春、河北、山东、广东、广西、浙江、韩国等地的NDV的同源性在96.3%~98.8%之间。并且这3株分离株F蛋白的裂解位点氨基酸组成为^112R-R-Q-K-R-F^117,具有典型的强毒株裂解位点的特点。 The fusion (F) protein genes of three newcastle disease virus (NDV) isolates from Inner Mongolia were amplified by RT-PCR and sequenced. The sequencing analysis showed that the F gene encoding 554 amino acids was 1 662 bp in length. The homologous compari- son of the three isolates showed 97.5%, 97.9% and 98.0% of identities for each other; 85.8% , 86. 4% and 86. 8% with F48E9 strain, re- spectively; 96. 3% -98.8% with the isolates from Changchun, Hebei, Shandong, Guangdong, Guangxi, Zhejiang and South Korea. The cleavage sites ( 112 - 117 ) of F protein from the three isolates were ,^112R-R-Q-K-R-F^117 , which is characteristic of the velogenic strain.
