摘要
采用DEAE Cellulose、DEAE SephadexA-50、Sephadex G-200三种不同层析柱通过柱层析路径1、2对广东虫草多糖(CGP)进行分离纯化。并利用凝胶渗透色谱法(GPC)测定各柱层析组分的保留时间tR、重均分子量Mw、多分散性PD以及各组分的相对含量,以表征不同柱层析方法所得多糖组分的分子量分布。结果表明:大分子量多糖组分CGP-Ⅰ经过柱层析路径1分离纯化后,分子量分布由宽变窄,分子量略有降低,多糖组分的纯度得到了很好地提高;CGP经过不同柱层析路径中的NaCl梯度洗脱后,得到的都是分子量分布较均匀的小分子多糖。GPC结果显示柱层析路径1对CGP的分离纯化效果优于路径2。
Three different chromatography column(DEAE Cellulose, DEAE Sephadex A-50, Sephadex G- 200)were used to purify Cordyceps guangdongensis polysaccharide(CGP) by column chromatography path 1 and path 2. Moreover, in order to characterize the molecular weight distribution of CGP fractions, retention time (t.), weight average molecular weight(Mw) polydispersity(PD) and relative content of CGP fractions were obtained by gel permeation chromatography (GPC). The results show that, after going through the column chromatography path 1, the molecular weight distribution of high molecular weight polysaccharide fraction CGP- I ,changed from broad to narrow, its molecular weight decreased somewhat, while the purity of polysaccharide fractions were upgraded; after undergoing NaCI gradient elution in different column chromatography paths, CGP was isolated to obtain small molecular polysaccharide fraction, which has even distribution of molecular weight. According to GPC results, the effect of column chromatography path 1 on the purification of CGP is better than that of path 2.
