温度对地塞米松所致下丘脑神经元[Ca^(2+)]_i下降的影响

摘要 目的:探讨不同温度对于地塞米松(DEX)介导的下丘脑神经元内游离钙离子浓度([Ca2+]i)下降幅度的影响。方法:使用原代培养的Wistar新生大鼠下丘脑神经元,用Fluo-3/AM钙荧光探针标记细胞内游离Ca2+,分别在18~19℃、21~22℃、24~25℃三种温度条件下,以10-6MDEX刺激D′hanks液中的神经元,在激光共聚焦显微镜下连续观察不同温度下神经元内钙荧光变化。结果:在24~25℃条件下,10-6MDEX刺激使培养的下丘脑神经元细胞内[Ca2+]i迅速减少20.6%;在21~22℃时减少约36.8%;而18~19℃时则减少约61.7%。结论:在一定范围内,DEX介导的下丘脑神经元[Ca2+]i下降幅度随着温度的降低而增大。 Objective:To investigate the effects of different temperatures on 10-6 M dexamethasone ( DEX)- induced decreases of [Ca^2+]i. L Methods : The hypothalamic neurons from 2 - 3 days old Wistar neonatal rats were primarily cultured, and then the intracellular Ca^2+ in these cells were marked with calcium fluorescent probe of Fluo - 3/AM. Under the temperature of 18 - 19℃ ,21 -22℃ ,24 - 25℃, 10 -6 M DEX was used to stimulate the hypothalamic neurons in D' hanks solution and the fluorescent values were recorded continuously under confocal laser scanning microscope. Results : Under 24 - 25℃, the exposure of 10 -6 M DEX induced 20.6% decrease of [Ca^2+]i in the cultured hypothalamic neurons within 2 seconds. Under 21 - 22 ℃ ,the DEX induced 36.8% decrease of [Ca^2+]i and under 18 - 19 ℃ ,61.7% decrease of [Ca^2+]i occurred. Conclusion:Within a certain range, the descending amplitude of [Ca^2+]i in the cultured hypothalamic neurons induced by 10-6 M DEX increases with the decrease of temperature.
