摘要
目的探讨自发性癫痫大鼠(SER)皮质和海马中GLT-1、EAAC1 mRNA与蛋白的表达情况。方法应用RT-PCR技术检测GLT-1、EAAC1 mRNA的表达;应用Westernblot方法检测GLT-1、EAAC1蛋白的表达;应用免疫组化方法定位GLT-1、EAAC1蛋白。结果SER皮质中GLT-1蛋白表达低于正常Wistar大鼠(P<0.05);EAAC1 mRNA及蛋白表达均低于正常Wistar大鼠(P<0.01);SER海马中GLT-1蛋白表达高于Wistar大鼠(P<0.01);免疫组化结果显示GLT-1、EAAC1在SER、Wistar大鼠皮质和海马中均有表达,且位于细胞膜上。结论SER皮质中GLT-1、EAAC1表达下调可能促进了癫痫的发生;海马中GLT-1蛋白表达上调可能是一种代偿性神经保护机制。
Aim To investigate mRNA and protein expression of GLT-1, EAAC1 in cortex and hippocampus of spontaneously epileptic rats (SERs)and Wistar rats served as a control study. Methods Total RNA was extracted from cortex and hippocampus of SER and Wistar rats, and GLT-1, EAAC1 mRNA expression were detected by RT-PCR; the expression of GLT-1, EAAC1 protein was revealed by Western blot and immunohistochemistry. Results GLT-1 protein of SERs decreased in cortex (P 〈 0. 05 ). EAAC1 mRNA and protein of SERs decreased significantly in cortex ( P 〈 0. 01 ). GLT-1 protein of SERs increased significantly in hippocampus(P 〈0.01 ). GLT-1, EAAC1 protein were expressed on the cellular membrane in both cortex and hippocampus of SERs and Wistar rats. Conclusions The expression of GLT-1, EAAC1 is down-regulated in SER cortex, whichmay contribute to the seizure; the expression of GLT-1 is up-regulated in SER hippocampus, which may be the compensatory response.
