摘要
用放射配基竞争结合分析测定连续激发哮喘模型大鼠和加味六君子汤治疗后不同时间肺组织GR的结合容量及血浆皮质酮含量(蛋白竟争结合法)。各哮喘组大鼠肺组织GR结合位点在激发后第1天显著升高;第3天迅速回落,其中中药组和中药加西药组仍维持较高水平,与正常对照组比较差异显著(P<001);第7天模型组继续下降,但与正常对照组比较差异不显著,与中药组、中药加西药组比较差异显著( P< 0. 05)。血浆皮质酮的含量,激发后从第 1天起各哮喘组均明显下降,与正常组比较差异显著(P<0.01);激发后第7天,中药组与模型组比较差异显著(P<0.01)。提示大鼠肺组织GR随哮喘发作的不同时期而变化,哮喘大鼠存在肾上腺皮质功能的不足,加味六君子汤可上调哮喘大鼠肺组织GR和提高内源性肾上腺皮质激素。
The binding capacity of glucocorticoid receptor (GR) of the lung tissue and plasmacorticosterone content were respectively measured by radioligand binding assay and competitiveprotein binding assay in different groups of asthmatic rats treated by supplemented Six Noble Decoction (SSND) and other drugs after continuous challenge. GR binding sites were markedly elevated in asthmatic groups 1 day after challenge and rapidly decreased on the thrid day, while theyremained in high level in rats treated with SSND or combined SSND and beclomethasone, beingsignificantly higher than those of the normal controls (P< 0, 01 ). Gr binding sites of the lung tissue in asthmatic model rats further declined on the seventh day after challenge, and though notstatistically different from those of the control rats, were signficantly different from those of SSND treated group and combined SSND and beclomethasone group(P < 0. 05 ). Plasma corticosterone contents in all groups of rats were significantly lowered ond day after challenge(comparedwith normal rats, P < 0. of ). Their contents in SSND group were statistically different fromthose of model rats (P < 0. 01 ). It is suggested that Gr binding capacity of the rat lung tissuechanges with the course of asthma attack and adrenocortical insufficiency is present in asthmaticrats. SSND can up - regulates GR of the lung tissue and elevates endogenous adrenocortical hormone content in these rats.
