摘要
目的:研究中药桃核承气改良方对家兔血管内皮损伤后STAT3(信号转导和转录活化因子)和VEGF(血管内皮生长因子)的影响。方法:健康新西兰大耳白兔40只,体重2~2.5kg,雌雄不拘。将动物分模型组(n=10)、中药组(n=10)、西药组(n=10)、空白组(n=10),模型组、中药组、西药组高酯饲料喂养4周后行腹主动脉内膜剥脱术造模。术后模型组、中药组、西药组正常饲料喂养,中药组予桃核承气改良方灌胃4周,西药组予舒降之、阿司匹林灌胃4周,模型组、空白组予生理盐水灌胃4周。采用半定量RT-PCR法测定血管中STAT3mRNA半定量和VEGF mRNA半定量。结果:模型组STAT3和VEGF mRNA半定量较空白组明显升高(P〈0.05);西药组STAT3和VEGF mRNA半定量较模型组明显降低(P〈0.05);中药组STAT3和VEGF mRNA半定量较模型组明显降低(P〈0.05),中药组与西药组之间无明显差异(P〉0.05)。结论:桃核承气改良方能防治血管内皮增生从而预防冠心病经皮冠状动脉介入治疗技术后冠脉再狭窄。
Ojective:To investigate the effects of improved proscription of Taoron Chengqi decoction (Peach Kernel decoction for strengthening qi) on the activation of STAT3 and VEGF of rabbits after intimadenudation. Methods :40 healthy New - Zealand white rabbits ( 2 - 2.5 kg) were divided into the model group ( n = 10), Chinese medicine group ( n = 10), western medicine group (n = 10 ), and the narmol group (n = 10 ). Before the operation, model group, Chinese medicine group and western medicine group were feeded with high fat diet for 4 weeks. Afer the operation, model group, Chinese medicine group and western medicine group were fecded with narmol diet for 4 weeks, and the intragastric administration of Chinese medicine group( n = 10) is the improved prescription of Taoron Chengqi decoction, western medicine group is simvastation and asprin. The expression of STAT3 and VEGF mRNA was determined using RT - PCR method. Results :The expression level of STAT3 and VEGF mRNA of model group are higher than the narmol group obviously. The expression level of STAT3 and VEGF mRNA of Chinese medicine group and western medicine group are decreased( Compared with the group of control, P 〈 0.05 ). The Chinese medicine group and western medicine group have no difference ( P 〉 0.05 ). Conclusion:The improved prescription of Taoren Chengqi decoction has an accelerating action on growth and function of vascular endothelial cell. It is effective in the prevention of restenosis after PCI.
