摘要 目的:观察不同脑缺血再灌注动物模型的脑损伤程度,探讨不同脑缺血再灌注模型的应用。方法:制备双侧颈总动脉阻断再灌模型(BCCO)、线栓法大脑中动脉脑缺血再灌模型(MCAO)、四血管法全脑缺血再灌模型(GI),采用海马CA1区MAP-2免疫组织化学染色观察其脑损伤情况,Morris水迷宫评价其行为学。结果:MAP-2免疫表达显示GI组脑损伤最严重,CA1区细胞数量最少,MAP-2表达最低,而BCCO组脑损伤最轻,CA1区细胞数量无明显减少,MAP-2表达仅略有降低。行为学研究表明,MCAO和GI组行为学明显改变。结论:不同的脑缺血模型的应用不同,BCCO模型可用于轻微缺血的预防,MCAO模型适用于局部损伤后脑功能改变及治疗药物评价,GI是研究缺血损伤后恢复相对理想的模型。 Objective: To investigate the application of three kinds of cerebral ischemia-reperfusion ani- mal model according to the degree of injury. Methods : The bilateral common carotid artery occlusion (BCCO) , cerebral middle cerebral artery occlusion (MCAO) by string-fastening method, global ischemia(GI) by four-artery occlusion were used to establish cerebral ischemia-reperfusion animal model. The degree of brain injury was evaluated through the expression of MAP-2 in hippocampal CA1 region and the Morris water maze. Results: The result of the expression of MAP-2 indicated the injury of GI group was the most severe, and the damage of BCCO group was the most mild. Compared with the normal group, the behavior change of reperfusion was the significant in the MCAO and GI groups. Conclusion :The three animal models should be applied in different researches. The BCCO model could be used in prevention of mild ischemia. The MCAO model could be used in the change of cerebral function and the drug evaluation. The GI model could be used in recovery after cerebral ischemia.
