Expression and significance of neurokinin B and placenta growth factor in placentas with hypertensive disorder complicating pregnancy

摘要 目的:探讨妊娠期高血压疾病(HDCP)孕妇胎盘组织中神经激肽B(NKB)、胎盘生长因子(PLGF)的表达及其与疾病发生发展的关系。方法:采用免疫组化法检测50例HDCP患者(妊娠期高血压患者15例,子癎前期轻度患者15例,子癎前期重度患者20例)及20例正常孕妇胎盘组织中NKB与PLGF蛋白定位及定量表达情况。结果:NKB蛋白及PLGF蛋白主要分布在绒毛合体滋养细胞和间质细胞的胞质,HDCP组胎盘NKB蛋白的表达高于正常组(平均灰度值分别为109.89±13.04和90.02±3.17,P<0.05),PLGF蛋白的表达低于正常组(平均灰度值分别为82.86±8.34和101.81±4.40,P<0.05);随着病情的加重,合体滋养细胞NKB的表达水平明显升高,PLGF表达水平明显降低(P<0.05)。结论:胎盘组织中NKB和PLGF蛋白的异常表达可能与HDCP的发生发展有关。 Aim : To determine the expression and significance of neurokinin B and placenta growth factor in placentas with hypertensive disorder complicating pregnancy. Methods:50 women with hypertensive disorder complicating pregnancy (15 gestational hypertension, 15 slight preeclampsia patients and 20 severe preeclampsia women )and 20 normal pregnant women were studied. The protein expression of NKB and PLGF was determined by immunohistochemistry. Results: The distribution of NKB and PLGF was in the cytoplasm of villous syncytiotrophoblast and villous stroma . The expression of NKB was significantly increased in hypertensive disorder complicating pregnancy placentas compared to the normal ones (the average gray scale was 109.89 ± 13.04 vs 90.02 ± 3.17, P 〈 0.05) , while PLGF level in the placenta from hypertensive disorder complicating pregnancy placentas was significantly decreased than the normal people ( the average gray scale was 82.86 ±8.34 vs 101.81 ±4.40, P 〈 0. 05 ) , and the expression of NKB in sycytiotrophoblast was significantly in- creased while PLGF was significantly decreased with the severity of I-IDCP ( P 〈 0.05). Conclusion : The abnormal expression of NKB and PLGF in placentas may play an important role in the pathogenesis of hypertensive disorder complicating pregnancy.
