摘要
本文对从吸水链霉菌17997发酵提取获得的格尔德霉素精制品进行了LC-MS-MS分析,发现其中含有4,5-双氢格尔德霉素组分。对吸水链霉菌17997摇瓶发酵产物的硅胶板薄层层析分析表明,4,5-双氢格尔德霉素组分在发酵中期有积累。已知4,5-双氢格尔德霉素的抗肿瘤活性比格尔德霉素差,因而需要限制其在格尔德霉素制品中的含量。本文研究结果对以吸水链霉菌17997等为产生菌研究格尔德霉素发酵,促进4,5-双氢格尔德霉素转化为格尔德霉素以提高发酵液的品质,以及检测格尔德霉素制品中的4,5-双氢格尔德霉素组分具有实际意义。
A minor component, preparation from Streptomyces hygroscopicus 4, 5-dihydrogeldanamycin, was discovered in the purified geldanamycin 17997 by LC-MS-MS. Analysis of the fermentation products of Streptomyces hygroscopicus 17997 by TLC of silica gel plate indicated that 4, 5-dihydrogeldanamycin appeared in visible amounts in the mid-stage of fermentation, when geldanamycin was biosynthesized in maximal speed. As the anti-cancer cell activities of 4, 5-dihydrogeldanamycin were far inferior to geldanamycin, the amounts of 4, 5-dihydrogeldanamycin should be kept to the lowest possible in geldanamycin preparations. The results obtained in this research could be applicable to the fermentation study of geldanamycin by Streptomyces hygroscopicus 17997, or other geldanamycin producers, to promote the transformation of 4, 5-dihydrogeldanamycin to geldanamycin, and the detection of 4, 5-dihydrogeldanamycin in quality control of geldanamycin preparation(s).
