摘要
目的探讨白细胞介素10(IL-10)基因表达及脾脏对肝再生过程的影响。方法通过大鼠70%肝切除制备肝再生模型,以流式细胞仪检测肝细胞DNA的复制、分子杂交显示IL-10的基因表达、免疫组化以及形态测量学方法了解IL-10在肝内的分布,经统计学分析判断肝损伤和切脾后IL-10对肝再生的影响。结果切除脾脏和CCl_4造成肝损害后,随DNA合成量的减少IL-10的表达增强;免疫组化检测IL-10阳性细胞获得了相同的结果。统计学相关分析表明IL-10与DNA的改变呈完全负相关。结论生理和肝损伤过程IL-10的基因表达均影响着肝脏的再生,脾脏对肝再生的调控部分是通过IL-10的基因表达发挥作用的。
Objective To study the effects of IL-10 gene expression in liver cells on liver regeneration after splenectomy or liver injury by CC1_4. Methods The rat model of liver regeneration were produced by partial hepatectomy. The hepatocytes DNA replication are tested using flow cytometry technique. The IL-10 gene expression in liver cells were showed by dot blot hybridization method. The synthesis and distribution of IL-10 protein in liver tissue were examined by immune histochemical and morphometric analysis methods. Results After splenectomy or liver injury by CC1_4, the increase of ILl0 gene expres-sion in liver cells was associated with the decrease of DNA replication, the same result was obtained in IL-10 positive cells detected by immune histochemical method. Statistical correlation analysis showed that the changes of IL-10 was completely negatively related to the changes in DNA. Conclusion The IL10 gene expression in liver ceils affected on the regeneration of hepatocytes, and the modulation of spleen on liver regeneration was partially acted through the IL-10 gene expression.
