摘要
目的探讨具有清热利湿、祛风通络作用的中药制剂悉通颗粒治疗痛风性关节炎的作用机制。方法32只雌性Wistar大鼠随机分为空白对照组、模型组、秋水仙碱组(阳性对照)、悉通颗粒组,以尿酸钠溶液注射大鼠踝关节造成痛风性关节炎模型,ELISA法检测大鼠TNF-α水平,免疫组化法观察大鼠关节软组织TGF-β1及TGF-βR1的表达。结果悉通颗粒组大鼠关节软组织TNF-α水平降低,TGFβ1及TGFβR1表达增加,与模型组比较有显著差异。结论悉通颗粒治疗痛风性关节炎的机制可能为通过抑制TNF-α致炎因子,上调TGF-β1及TGF-βR1的适度表达。
Objective: To explore the therapeutic mechanism of Xitong Granule in treating gouty arthritis. Methods: 32 female Wistar rats were randomly divided into four groups: control, model, colchieine and Xitong Granule group. The monosodium urate was injected into ankle joint cavity for gouty arthritis model establishment. The level of TNF - α was measured by ELISA. The expression of TGFβ1 and TGFβR1 were examined by immunohistochemistryt. Results: The level of TNF - α in soft tissue surrounding ankle joint in Xitong Granule group was lower than those in model group, but the expression of TGFβ1 and TGFβR1 was significantly increased. Conclusion: The mechanisms of Xitong Granule treating gouty arthritis may be through inhibiting levels of TNF - α and up - regulating the expression of TGFβ1 and TGFβR1 of soft tissue surrounding ankle joint.
