摘要
目的:检测DNA甲基转移酶-1(DNMT-1)在特应性皮炎(AD)患者中的表达。方法:应用逆转录酶-聚合酶链反应(RT-PCR)结合凝胶图像扫描技术检测培养外周血PBMC的DNMT-1基因。采用电化学发光-酶联免疫测定法检测血清总IgE。结果:AD患者组与正常人对照组DNMT-1基因表达无显著差异(P=0.126)。AD患者中血清IgE水平高于5000 IU/mL者的DNMT-1基因表达显著低于正常对照组(P=0.0033),其他组间的表达无差异。DNMT-1基因表达水平与血清IgE水平没有明显的相关性。结论:外周血DNMT-1基因表达水平低下可能与血清IgE水平较高的AD的发病机制有关。
Objective: To measure the expression of DNA methyltransferase- 1 (DNMT- 1 )in the patients with atopic dermatitis (AD). Method: The expression of DNMT - 1 gene of peripheral blood mononuclear cells (PBMC) isolated was detected by reverse transcriptase - polymerase chain reaction (RT - PCR) with gel image scanning. The total serum IgE level was measured by chemiluminescent enzyme immunoassay. Results: There was no significant difference in the level of DNMT - 1 mRNA between AD patients and controls ( P = 0.126). The level of DNMT - 1 mRNA in the AD patients with IgE level 〉 5000 IU/mL was lower than that in normal controls ( P = 0.0033). The expression of DNMT- 1 mRNA between other groups was not significantly different. In addition, there was no signif-icant relationship between the expression of DNMT- 1 mRNA and IgE level in the current study. Conclusion: The observation suggests that suppression of DNMT - 1 may be related to the pathogenesis of AD, particularly in those whose serum IgE level is high.
