摘要
目的检测系统性红斑狼疮(SLE)患者血清亲环素A(CyPA)水平,探讨CyPA参与SLE活动的病因机制。方法应用ELISA检测57例SLE患者和30例正常人血清CyPA水平;应用重组人CyPA(rhCyPA)和/或植物血凝素(PHA)刺激培养Jurkat细胞株,24h后收集细胞上清,再用ELISA检测上清中干扰素γ(IFN-γ)的含量。结果SLE活动期患者血清CyPA水平明显高于稳定期患者及正常人(P<0.05);Jurkat细胞株仅在rhCyPA和PHA共刺激下,IFN-γ的分泌才明显增加(P<0.05)。结论rhCyPA和PHA能协同刺激Jurkat细胞株分泌IFN-γ;SLE患者外周血CyPA异常升高可能参与刺激T细胞分泌IFN-γ,促进SLE活动,可作为评价SLE活动的参考指标。
Objective To investigate the association of systemic lupus erythematosus (SLE) activity and cyelophilin A (CyPA) level in peripheral blood. Methods Serum levels of CyPA were detected by ELISA in 57 SLE patients and 30 normal controls. Jurkat cell lines were cultured and stimulated with reconstructive human CyPA(rhCyPA), PHA or rhCyPA plus PHA, respectively. Culture supernatants were collected, for detecting cytokine interferon factor-γ(IFN-γ) by ELISA. Results The CyPA levels were signifcantly higer in the active SLE subjects than those in the inactive SLE or normal controls (P〈0. 05). The IFN-γ level in culture supernatant secreted by Jurkat cell lines was increased only after stimulated by rhCyPA plus PHA at same time. Conclusion rhCyPA and PHA can synergistically induce Jurkat cell lines secreting IFN-7. Abnormal increase of CyPA in peripheral blood of SLE patients may participate in stimulating T cells secreting IFN-γ and activating SLE,which may be taken as an useful clinical index for evaluating SLE activity.
