摘要
目的:建立心脑康胶囊中葛根素和2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定的RP-HPLC方法。方法:色谱柱:Nova-Pak C_(18)(300mm×3.9mm,4μm),流动相:乙腈-水(20:80),流速:0.8 ml·ml^(-1),检测波长:320nm。结果:葛根素的线性范围为0.31~1.53mg·ml^(-1)(r=0.999 8),平均回收率为98.67%,RSD=1.24%(n=6);2,3,5,4′-四羟基二苯乙烯-2- O-β-D-葡萄糖苷的线性范围为1.21μg·ml^(-1)~9.65μg·ml^(-1)(r=0.999 6),平均回收率为98.43%,RSD=1.02%(n=6)。结论:方法简便、快速、准确,适合该胶囊中葛根素和2,3,5,4′-四羟基二苯乙烯-2-O-β-D-葡萄糖苷的含量测定。
To establish a method to determine the contents of puerarin and 2,3,5,4'-tetrahydroxy-stibene-2-O-β-D-glycoside in Xinnaokang capsule by RP-HPLC. Method: Nova-Pak C^s column(300mm×3.9mm,4μm) wsa used, the acetonitrile-water(20: 80) was used as mobile phase, the flow rate was 0.8mg·ml^-1, and detection wavelength was at 320nm. Result: The linear range of puerarin was 0.31 - 1.53mg·ml^-1 ( r = 0. 999 8 ) , the average recovery was 98.67% with RSD of 1.24% ( n = 6 ). and The linear range of 2,3,5,4'-Tetrahydroxy-stibene-2-O-β-D-glycoside was 1.21 -9.65μg·ml^-1( r = 0. 999 6), the average recovery was 98.43% with RSD of 1.02% (n = 6). Conclusion: The method is simple, sensitive and accurate. It can be used for the determination of puerarin and 2,3,5,4'-Tetrahydroxy-stibene-2-O-β-D-glycoside in Xinnaokang capsule.
