百草枯中毒大鼠肾损伤模型的建立 被引量:6

Establishment of paraquart-induced renal injury model in rats

摘要 目的观察并分析不同染毒剂量百草枯(PQ)中毒大鼠肾组织病理表现,寻找观察百草枯中毒大鼠肾损伤的最佳染毒剂量。方法SD大鼠腹腔注射不同剂量PQ,观察不同时间点肾组织病理表现,并计算实验大鼠的死亡率。结果各染毒组病理损伤在染毒后第1d即出现,以皮质部近曲小管受累最常,同一染毒组在不同时间点病理损伤的严重程度无明显差别。15mg/kg和20mg/kg染毒组可见皮质部小管上皮细胞肿胀,部分空泡变性;35mg/kg染毒组皮质部小管上皮细胞细胞核固缩增多,上皮细胞结构消失,少数肾小球结构紊乱,髓质部小管细胞核固缩,管腔消失。25mg/kg和30mg/kg染毒组病理损伤程度介于上述两组之间。随染毒剂量增加,实验大鼠病死率升高;25mg/kg染毒组死亡率为48.7%、30mg/kg染毒组死亡率为68.4%。结论25mg/kg染毒剂量可能是观察百草枯中毒大鼠肾损伤的最佳染毒剂量。 Objective To observe pathologically the toxic effect of paraquat (PQ)at various doses on kidney of rats Methods SD rats were exposured to PQ at various doses intraperitoneally, and pathological changes of kidney were observed at different time points. Results First day after PQ treatment,renal pathological changes could be seen in all groups, proximal tubule at cortical most serevely involved. No significant differences could be found at different time point for each PQ dose group. In 15mg/kg and 20mg/kg, dose groups ,tubular epithelial cells of cortical showed swelling,part of which showed vacuole degeneration. In 35mg/kg dose group, the karyopyknosis of the tubular epithelial cells of cortical increased. The structure of epithelium disappeared, that of glomcndus disorganized and medulla were involved. In 25mg/kg and 30mg/kg dose groups , severity of pathological changes were between that of 20mg/kg and 35mg/kg dose groups. The rats mortality rate was increased along with the increase of the dosage too, being 48.7% in 25mg/kg dose group ahd 68.4% in 30mg/kg dose group. Conclusion 25mg/kg is the optimum does for observation of paraquart-induced renal injury in rats.
