摘要
目的:建立HPLC法测定广西红腺忍冬中绿原酸含量。方法:采用Hypersil ODS-2柱(4.6mm×250mm,5μm),以甲醇-乙腈-0.05%磷酸为流动相,梯度洗脱,流速0.5mL·min-1,检测波长238nm,柱温30℃。结果:对不同产地、不同加工方法的23个样品进行了绿原酸含量测定,绿原酸进样量在0.2~1.0μg范围内呈良好线性关系(r=0.9999),平均回收率(n=6)为100.5%(RSD=2.8%)。结论:不同产地及不同加工方法的样品中绿原酸含量差别较大,各样品绿原酸含量均远高于中国药典要求。
Objective:To establish an RP - HPLC method for determination of chlorogenic acid in Flos Lonieerae Hypoglaucae from Gnangxi. Method : Hypersil ODS - 2 (4.6 mm×250 mm,5 μm) was adopted ; The mobile phase was methanol - acetonitrile - 0.05 % phosphoric acid with gradient elution, and the flow rate was 0.5 mL·min^-1; The detective wavelength was 238 nm and column temperature was 30℃. Results: Chlorogenic acid was determined on 23 samples of Flos Lonieerae Hypoglaucae from different producing areas and processing methods. The linear range of ehlorogenic acid was 0.2 - 1.0 μg( r = 0. 9999 ) ;The average recovery( n = 6) was 100.5% ( RSD = 2. 8% ). Conclusion:The difference between the content of chlorogenic acid in different samples with different origins, different processing methods was marked. The content of chlorogenic acid in all samples is far higher than the standard of Chinese Pharmacopoeia.
