摘要
目的:建立枸杞叶中芦丁的含量测定方法。方法:采用Diamonsil C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.5%磷酸溶液,梯度洗脱,流速为1.0mL·min-1,检测波长为358nm。结果:芦丁进样浓度在50.7~404.8μg·mL-1范围内线性关系良好(r=0.9999);低、中、高浓度的平均回收率(n=3)分别为101.8%(RSD=1.8%),98.6%(RSD=1.5%),97.9%(RSD=0.7%)。结论:本法准确、可靠,灵敏度高,可作为枸杞叶中芦丁的定量分析方法。
Objective: To develop an HPLC method for the determination of rutin in Lycium barbarum leaves. Method:A Diamonsil- C18(250 mm ×4.6 ram,5 μm) column was adopted. The mobile phase was a mixture of methanol - 0.5 % phosphoric acid solution with gradient elution program. The flow rate was 1.0 mL·min^-1. UV detection wavelength was at 358 nm. Result: A good linearity was obtained in the range of 50.7 - 404.8μg·mL^-1 for rutin. The average recoveries(n = 3 ) of low, middle, high concentration were 101.8% (RSD = 1.8% ) ,98.6% ( RSD = 1.5% ) and 97.9% ( RSD = 0. 7% ), respectively. Conclusion: This method is simple, accurate, sensitive and suitable for the determination of rutin in Lycium barbarum leaves.
