摘要 目的探讨P物质及其受体神经激肽1(NK-1)在体外培养的乳癌细胞系T47D中的表达及免疫细胞化学定位。方法用放入盖玻片的6孔板进行细胞培养,第3天将盖玻片取出进行免疫细胞化学染色。结果在T47D细胞系中P物质和NK-1呈阳性表达。P物质阳性表达主要位于胞浆,而NK-1阳性表达主要位于胞膜和胞浆。结论P物质和NK-1在绝大多数T47D细胞中呈阳性表达,提示神经内分泌机制参与了乳癌的发病过程。该研究揭示了T47D细胞系的一种新特性,并为乳癌的治疗提供了新的可能靶点。 Objective To investigate the immunolocation of substance P and NK-1 in breast cancer cell line T47D. Methods All cells were cultured in 6-well tissue-culture plates with cover slips in. The cover slips were removed from the plates on the third day, and immunocytochemical stain done. Results Both substance P and NK-1 were positively expressed in T47D cells. Substance P was mostly localized within the cytoplasm, and NK-1 expressed mainly in both cell membrane and/or cytoplasm. Conclusion The substance P and NK 1 show positive expression in most T47D cells, which indicate that neuroendocrine mechanism involves in breast cancer development. This study reveals a new characteristic of T47D cell line an offers a new possible target for breast cancer therapy.
