摘要
目的:说明丹参酮ⅡA对血管合成血管紧张素Ⅱ及醛固酮的作用。方法:用大鼠肠系膜动脉离体灌注、高效液相色谱法和放射免疫法检测肠系膜动脉中合成的血管紧张素Ⅱ及醛固酮。用逆转录聚合酶链反应(RT-PCR)及SouthernBlot法检测血管中醛固酮合酶基因CYP11B2信使核糖核酸(mRNA)的表达情况。结果:丹参酮ⅡA抑制血管合成醛固酮,但不能抑制血管合成血管紧张素Ⅱ。丹参酮ⅡA抑制血管中醛固酮合酶基因CYP11B2mRNA的表达。结论:丹参酮ⅡA治疗心血管疾病的机制可能与其抑制血管合成醛固酮有关。
Objective:To find the pharmacological effects of tanshinone ⅡA on angiotensin Ⅱ and aldosterone production in blood vessels. Methods:Using ex vivo rat mesenteric artery perfusion,HPLC,and RIA to measure the amount of angiotensin Ⅱ and aldosterone produced in vascular tissue,and by RT PCR and Southern Blot to detect the expression of aldosterone synthase gene—CYP11B2 mRNA in aorta. Results:The production of aldosterone but not of angiotensin in mesenteric artery was inhibited ( p <0.05) and the CYP11B2 mRNA expression in aorta was down regulated by tanshinone ⅡA. Conclusion:The inhibition of vascular aldosterone by tanshinone ⅡA might contribute to its therapeutic effect on cardiovascular diseases.
