摘要
利用SRAP分子标记从700对引物中筛选到普通烟草栽培种内(Nicotiana tabacum L.)两条晾烟(马里兰烟、白肋烟)类型的特征标记带、一条晒烟(香料烟和名优晒烟)类型的特征标记带,一条烤烟与晾烟特征标记带。这些标记不但可以直接转化成SCAR标记,而且可通过分离群体的遗传分析用于烟草类型间遗传转化的特征带,也可进一步用于测序发掘新基因。在分子水平上为研究普通烟草种内不同栽培类型间的遗传差异和类型划分奠定了可行性实验基础。
In this study, four characteristic bands for distinguishing cultivated forms of Nicotiana tabacum L. were screened by 700 pairs of SRAP primers. Among the four characteristic bands, air-cured tobacco including maryland tobacco and Burley tobacco,sun cured tobacco including oriental tobacco and sun-cured tobacco, and flue-cured tobacco and air-cured tobacco were distinguished. The four bands can be used either directly as the characteristic bands of in genetic transformation among cultivated forms of Nicotiana tabacum L. , or as SCAR markers for further study. Furthermore, the characteristic bands could be sequenced and the corresponding genes of which could be isolated in the future research. Therefore, the results provided a feasible research foundation for exploring the genetic diversity of different cultivated forms and the division of Nicotiarra tabacum L. at molecular level.
