无机氟对大小鼠胚胎中脑细胞增殖和分化的影响 被引量:8

EFFECT OF FLUORIDE PROLIFERATION AND DIFFERENTIATION IN RAT AND MOUSE EMBRYO MIDBRAIN CELL IN VITRO

摘要 选用SD大鼠,昆明种小鼠,采用胚胎中脑细胞微团培养方法,观察无机氟对胚胎中脑细胞增殖和分化的影响。结果显示培养液中一定浓度的氟对两种动物胚胎中脑细胞增殖和分化有明显抑制作用,呈剂量—反应关系。拟合细胞分化和增殖抑制曲线,计算大、小鼠半数抑制分化浓度(ID_(50))分别为7.1μg/ml、8.5μg/ml;半数抑制增殖浓度(IP_(50))为40.3μg/ml、51.5μg/ml IP_(50)/ID_(50)比值分别为5.68和6.06。表明无机氟是中脑细胞分化的特异性抑制剂,可能是潜在的致畸性。并表现有一定的种属差异. The effect of fluoride on proliferation and differentiation of rat and mouse embryo midbrain cells were studied with micromass cultures in vitro. Embryo midbrain cells of ral (13 - day) and mouse (12 - day) were used to culture for 5 days. The results showed that fluoride could inhibit differentiation of cells without affecting cells proliferation. The concentration of fluoride inhibition cell differentiation (ID50) were 7 .1 μg/ml (rat) , 8.5 μg/ ml (mouse) respectively. The concentration of inhibition cell proliferation(IP50)were 40.3μg/ml,51. 1 μg/ml assessment criteria of Flint and Cheng Wanrong,the fluoride should be embryo midbrain cells specific differentiation inhibitor. It could have potent teralogenecity.
