磁性载体用于酶固定化方面的研究 被引量:5

IMMOBILIZATION OF A αAMYLASE ONTO A MAGNETIC COLLIDAL PARTICLE BY ADSORBING-CROSSLINKING

摘要 合成了具有磁响应性的两亲性聚乙二醇胶体粒子,并以此为载体,用吸附交联法固定化了α淀粉酶.最适交联条件研究表明:缓冲液pH值、戊二醛浓度及加酶量都对固定化酶活力、比活有一定影响.在最适固定化条件下,固定化酶的活力为34000U·g-1干胶,蛋白载量为100mg·g-1干胶,比活为340U·mg-1蛋白,活性回收率为37.7%.最适反应温度比天然酶(50℃)提高30℃.最适pH比天然酶(7.0)提高1个pH单位.并且对酸、碱、热的稳定性大大增强,贮存及操作稳定性也大大提高.该载体合成简单,固定化方法简单,且具有磁性,便于分离. A magnetic immobilized αamylase has been prepared by adsorbing αamylase onto a magnetic collidal particle and crosslinking it with glutaraldehyde.The effects of different concentration of glutaraldehyde,pH of buffer,and the amout of αamylase for immobilization and the properties of magnetic immobilization αamylase were studied.The results showed that the activity of MIE was 34000U·g-1dry gel,the protein binding capacity was 100mg·g-1dry gel,the activity rentention was 37.7%.The optimum temperature of MIE was 30 degree higher than that of natural αamylas(50℃).The optimum pH values of MIE was 1pH values higher than that of natural αamylase(7.0).The magnetic immobilized αamylase has excellent thermal stability,pH stability and working stability.
