以苹果砧木平邑甜茶(Malus hupehensisRehd)和M7(Malus pumila Var.ParadisiacaSchneid)的组培苗叶片为材料,研究了暗培养、接种方式和植物生长调节剂对叶片不定芽再生的影响。结果表明,适合二者叶片不定芽再生的最佳暗培养时间是14d。平邑甜茶叶片以远轴面向下接触培养基再生效果好,叶片接种方式对M7叶片不定芽再生无显著影响。适合平邑甜茶叶片不定芽再生的最佳培养基是MS+6-BA2.0mg/L+IBA0.1mg/L,再生率达93.3%。适合M7叶片不定芽再生的最佳培养基是MS+6-BA4.0mg/L+IBA0.3mg/L,再生率达96.7%。适合平邑甜茶新梢伸长的最佳培养基是MS+6-BA0.3mg/L+IBA0.03mg/L+GA30.5mg/L,适合M7新梢伸长的最佳培养基是MS+6-BA0.5 mg/L+IBA0.05mg/L+GA31.0mg/L。适合平邑甜茶生根的最佳培养基为1/2MS+IBA0.1mg/L,生根率为86.7%。适合M7生根的最佳培养基是1/2MS+IBA1.0mg/L,生根率为93.3%。
The effects of dark treatment, inoculation methods and plant growth regulators on adventitious bud regeneration from leaves in vitro of Malus hupehensis Rehd and M7 ( Malus pumila Var. Paradisiaca Schneid) were studied. The results indicated that 14 days of dark treatment could effectively stimulate the adventitious bud regeneration from leaves of Malus hupehensis Rehd and M7. Higher regeneration rate of Malus hupehensis Rehd was found when the abaxial layer of leaves was contacted with the medium. However, inoculation methods of leaves had no significant effect on adventitious bud regeneration of M7. The most suitable medium for adventitious bud regeneration ofMalus hupehensis Rehd and M7 were MS +6 - BA 2. Omg/L + IBA 0. lmg/L and MS +6 - BA 4.0mg/L + IBA 0.3mg/L respectively. The regeneration rates of adventitious bud of Malus hupehensis Rehd and M7 were 93.3% and 96.7% respectively. The appropriate medium for plantlet elongation ofMalus hupehensis Rehd and M7 were MS + 6 - BA 0.3mg/L + IBA 0.03 mg/L + GAa 0.5mg/L and MS + 6 - BA 0.5 mg/L + IBA 0.05 mg/L + GA3 1.0 mg/L respectively. The best medium for roots induction ofMalus hupehensis Rehd and M7 were 1/2MS + IBA 0.1 mg/L and 1/2MS + IBA 1.0 mg/L respectively, and the rooting rates ofMalus hupehensis Rehd and M7 were 86.7% and 93.3% respectively.
