肺癌XPD基因表达与微卫星不稳定关系的探讨

Association between expression of XPD and microsatellite instability in lung cancer

摘要 目的:探讨肺癌中XPD基因表达及其与微卫星不稳定(MSI)的关联情况。方法:采用微卫星分析和RT-PCR方法检测了5个微卫星标记在肺癌中的状况及XPD转录水平的表达。结果:27.16%(22/81)的肺癌出现至少1个微卫星的不稳定,但是XPD没有明显的mRNA表达下调。MSI与肺癌分期、分级、淋巴结转移和患者性别等参数之间无相关性,P>0.05。结论:XPD基因微卫星的不稳定是肺癌的频发事件,与XPD基因表达可能不存在必然联系。 OBJECTIVE: To investigate microsatellite instability (MSI) of XPD in lung cancer and estimate the correlation between MSI and the expression of XPD gene. METHODS: MSI of 5 microsatellite markers and mRNA expression of XPD were assessed in a large cohort of patients with lung cancer by using denaturing polyaerylaide gel electrophoresis (PAGE) and reverse transcriptase-polymerase chain reaction (RT-PCR) techniques. RESULTS: A total of 27.16% (22/81) of tumors displayed MSI in at least one tested marker. No down-expression of XPD was found at the transcriptional level. The presence of XPD MSI did not correlate with the tumor stage, degree of differentiation, lymphonode metastasis, and sex of the patients(P〉0.05). CONCLUSION: MSI of XPD is common in lung cancer, but is probably not associated with the expression of XPD gene.
